    Cytogenetic Analysis and Clinical Phenotype of Primary Amenorrhea in Indonesian Patients (HASIL TURNITIN)

    Background: Primary amenorrhea (PA) is a symptom that can be caused by different disorders such as gonadal, endocrinal, physiological and genetic disorders.  Aim of study: This study provided the clinical and cytogenetic profiles of Indonesian primary amenorrhea patients and introduced clinical criteria of those patients with their  karyotype results using score system.   Methods: A retrospective descriptive study of 79 PA patients, whom referred to Cytogenetic and Molecular unit Center for Biomedical Research (CEBIOR), Faculty of Medicine Diponegoro University. We made a scoring system consisted of 4 scores, all patients had been distributed to match the scores according to their clinical criterias and then confirmed with the karyotype results.   Results: The karyotype results of 79 patients of PA revealed 55 (69.6%) patients with female karyotype 46,XX; 6 (7.6%) patients with male karyotype 46,XY; 8(10.1%) patients with monosomy X; 3 (3.8%) patients with 45,X/46,XX; 3 (3.8%) patients with  Isochromosome 45 X/46, X,i(Xq). Mosaicism with  Y  constitution 45,X/46,XY  was seen in 2 (2.5%) patients;  marker chromosome 45,X/46,X+mar (2%) in 1 patient (1.3%); and  chromosome 1 and  X translocation 46,XX,t(1;X)(p34;q25) detected in  1(1.3%) patient. Scoring system results showed that all patients with normal karyotype (46,XX/46,XY) were matched with score 1 and 2 while 17 patients with chromosomal abnormalities were matched with score 3 and 4, only 1 patient with mosaic Turner syndrome 45,X(10%)/46,XX(90%) matched score 1.  Conclusion: Turner syndrome was the most common cause of primary amenorrhea which attests the importance of cytogenetic analysis for diagnosis of primary amenorrhea patients. The scoring system needs further validated for measuring reliability and validity

    Human papilloma virus infection: vaccine knowledge attitude, and informational behavior among undergraduate students

    Background: The study aimed to know the Human Papilloma Virus infection, vaccine knowledge attitude, and informational behavior among university students. Methodology: The study was conducted on students of Government College University Faisalabad from January 2022 to March 2022. The study consisted of a questionnaire that verbalized the awareness and prevalence of HPV in particular age groups male and female students. The questionnaire was divided into four sections to assess the awareness, knowledge, and behavior of students toward the Human Papilloma Virus (HPV). Results: In the demographic representation about 85% (n=235 out of 275) of students belong to the age group of 18-22 while 14% (n=38 out of 275) students were from 23 to 24 years old. 23%(n=64) students were males while 76%(n=211) participants were females. Figures collected showed that 1%(n=3) participants were married while the remaining (about 98%) were single. According to the figures, about 70% of the partakers knew about the mode of transmission and the complications caused by the Human Papilloma Virus while 34% of all the students had no idea about HPV, its transmission, complications, or treatment. 65% had an idea that HPV causes genital warts and 75% were aware of the HPV vaccine. Moreover, 65% of students thought condoms as a source of prevention against HPV and 54% marked contact with body fluids as a mode of transmission of HPV. Conclusion: The knowledge and attitude towards HPV infection and HPV-related diseases and HPV vaccine among female students in the Government College University Faisalabad were significant. There is a need of well-defined education programs in the form of seminars, workshops and symposiums on the awareness of HPV infection are necessary to fill the knowledge gap among general population

    Pharmacological evaluation of Vernonia elaeagnifolia (Asteraceae) leaves in hyperlipidemic albino rabbits

    Purpose: To evaluate the antihyperlipidemic efficacy and phytochemical constituents of Vernonia elaeagnifolia aqueous leaf extract.Method: Qualitative phytochemical analysis of V. elaeagnifolia leaves was performed. Thirty healthy albino rabbits were divided into six groups (n = 6). Cholesterol powder (0.5 g/kg) in 10 mL coconut oil (vehicle) was given orally to induce hyperlipidemia. The aqueous leaf extract of Vernonia elaeagnifolia was administered at 250 mg/kg and 500 mg/kg per oral. Lipid profile, hepatic enzymes and oxidative stress markers were evaluated.Results: Phytochemical screening indicated the presence of tannins, proteins, flavonoids, phenols, alkaloids and saponins. Oral administration of cholesterol powder significantly (p < 0.05) raised the level of serum total cholesterol (TC), low-density lipoprotein cholesterol (LDL-c) and triglyceride (TG) along with significant (p < 0.05) decrease in serum concentration of high-density lipoprotein cholesterol (HDLc). Concentration of serum TC, LDL-c, TG and liver enzymes was significantly reduced in V. elaeagnifolia-treated groups. The levels of oxidative stress markers were restored to normal when the animals were treated with V. elaeagnifolia leaf extract; increased levels of antioxidant enzymes were observed.Conclusion: The aqueous leaf extract of V. elaeagnifolia possesses antihyperlipidemic and antioxidant potentials that are dose-dependent.     Expression of the androgen receptor, pAkt, and pPTEN in breast cancer and their potential in prognostication

    BACKGROUND: Importance of androgen receptor (AR) as an independent prognostic marker in Pakistani women with breast cancer (BCa) remains unexplored. Our aim was to identify the expression and potential prognostic value of AR, its upstream regulator (pAkt) and target gene (pPTEN) in invasive BCa. METHODS: This study used a cohort of 200 Pakistani women with invasive BCa diagnosed during 2002-2011. Expression of AR, pAkt and pPTEN was determined on formalin fixed paraffin embedded tissue sections by immunohistochemistry. The association of AR, pAkt and pPTEN with clinicopathological parameters was determined. Survival analyses were undertaken on patients with ≥ 5 years of follow-up (n = 82). RESULTS: Expression of AR, pAkt and pPTEN was observed in 47.5%, 81.3% and 50.6% of patients, respectively. AR-expressing tumors were low or intermediate in grade (P \u3c .001) and expressed ER (P = .002) and PR (P = .001). Patients with AR+ tumors had significantly higher OS (Mean OS = 10.2 ± 0.465 years) compared to patients with AR− tumors (Mean OS = 5.8 ± 0.348 years) (P = .047). Furthermore, AR-positivity was associated with improved OS in patients receiving endocrine therapy (P = .020). Patients with AR+ /pAkt+ /pPTEN− tumors, had increased OS (Mean OS = 7.1 ± 0.535 years) compared to patients with AR−/pAkt+/pPTEN− tumors (Mean OS = 5.1 ± 0.738 years). CONCLUSION: AR-expressing tumors are frequently characterized by low or intermediate grade tumors, expressing ER and PR. In addition, expression of AR, pAkt and pPTEN, could be considered in prognostication of patients with invasive BCa

    Cytokine changes in colonic mucosa associated with Blastocystis spp. subtypes 1 and 3 in diarrhoea-predominant irritable bowel syndrome

    We determined cytokines (e.g. interleukin-8, 10, 12 and TNF-α) expression by peripheral blood mononuclear cells (PBMCs) and in rectal mucosa in diarrhoea-predominant irritable bowel syndrome (D-IBS) with Blastocystis spp. Eighty patients with D-IBS and Blastocystis spp. infection were classified as \u27cases\u27 and 80 with D-IBS without Blastocystis spp. infection were classified as \u27control\u27. Cases were subdivided into D-IBS and Blastocystis sp. defined type 1 (subtype-specific primer SB83) and type 3 (SB227). Stool microscopy and culture were performed. Rectal biopsies were obtained for histology and cytokines by real-time PCR for mRNA expression of cytokines. PBMCs IL-8 was similar in different groups but in type 1, IL-8mRNA was increased compared with type 3 (P = 0·001) and control (P = 0·001). In type 1, IL-10 by PBMCs had a low mean value (14·5±1·6) compared with (16·7±1·5) type 3 and (16±2·3) in controls (

    The Pakistan risk of myocardial infarction study: A resource for the study of genetic, lifestyle and other determinants of myocardial infarction in south Asia

    The burden of coronary heart disease (CHD) is increasing at a greater rate in South Asia than in any other region globally, but there is little direct evidence about its determinants. The Pakistan Risk of Myocardial Infarction Study (PROMIS) is an epidemiological resource to enable reliable study of genetic, lifestyle and other determinants of CHD in South Asia. By March 2009, PROMIS had recruited over 5,000 cases of first-ever confirmed acute myocardial infarction (MI) and over 5,000 matched controls aged 30-80 years. For each participant, information has been recorded on demographic factors, lifestyle, medical and family history, anthropometry, and a 12-lead electrocardiogram. A range of biological samples has been collected and stored, including DNA, plasma, serum and whole blood. During its next stage, the study aims to expand recruitment to achieve a total of about 20,000 cases and about 20,000 controls, and, in subsets of participants, to enrich the resource by collection of monocytes, establishment of lymphoblastoid cell lines, and by resurveying participants. Measurements in progress include profiling of candidate biochemical factors, assay of 45,000 variants in 2,100 candidate genes, and a genomewide association scan of over 650,000 genetic markers. We have established a large epidemiological resource for CHD in South Asia. In parallel with its further expansion and enrichment, the PROMIS resource will be systematically harvested to help identify and evaluate genetic and other determinants of MI in South Asia. Findings from this study should advance scientific understanding and inform regionally appropriate disease prevention and control strategies
